BES is a zwitterionic buffering agent used in biochemistry and molecular biology that was first selected and described by Good et al. It is a bis(2-hydroxyethyl)amine buffer that is useful for a pH range of 6.4 – 7.8. It is soluble in water at 0°C up to 3.2M.

- A buffer is an aqueous solution consisting of a mixture of a weak acid and its salt (acidic buffer) or a weak base and its salt (basic buffer) that resists a change in pH on the addition of either acid or base.
- Its pH changes very little when a small amount of strong acid or base is added to it and thus it is used to prevent changes in the pH of a solution.

The pKa of the buffer should be near the desired midpoint pH of the solution. (2) The capacity of a buffer should fall within one to two pH units above or below the desired pH values. If the pH is expected to drop during the procedure, choose a buffer with a pKa slightly lower than the midpoint pH.
